Mechanism of Action Of Lorenol®

Suppression of Activation of Inflammatory Genes



  1. Chemicals, allergens, UVR events, and even irritants can cause a “triggering” event that causes keratinocytes and fibroblasts to produce inflammatory mediators, such as cytokines IL, prostaglandin PGE-2, and TNF-alpha, as well as the chemokines, MCP-1, and IL -8.


  1. Inflammatory Mediators such as these can travel to other cells in the skin, including other keratinocytes, pigment cells (melanocytes), other fibroblasts, and even to the cells that make up the blood vessels.


  1. As the Inflammatory Mediators reach each cell, they bind to the receptor embedded in the target cell's membrane ( see the diagram showing how the mediators bind to their particular receptor. Each mediator has a unique structure that allows it to recognize its particular receptor.


  1. Inflammatory mediators bind and “activate” the receptorThe receptor then creates an intracellular signaling pathway that ultimately reaches the nucleus and activates genes that cause inflammation and skin damage (e.g., genes for MMPs, genes for other cytokines, genes that shut off cell growth, etc.) Note that there are different signaling pathways (or different mediators and note how complicated and how many steps are involved in the “signaling pathways.


  1. The combination of ingredients in LORENOL® inhibits the various signaling pathways, in turn preventing a mediator from becoming activated and causing inflammation.

LORENOL® addresses "triggers" of Inflammatory Mediators such as:


  • TNF alpha - Is responsible for inflammatory skin issues such as atopic dermatitis, psoriasis, and rosacea, etc.


  • LTB-4 Luekotrien B4-Is responsible for asthma and inflammatory cytokines (Erythema and Edema)


The Interleukin-1 family (IL-1 family) is a group of 11 cytokines that play a central role in the regulation of immune and inflammatory responses to infections or sterile insults.


  • IL-1(interleukin-1) - Promotes aging, turns off collagen, elastin, and hyaluronic acid, and slows the growth rate. Stimulates the production of mmp’s.


  • IL-8 Responsible for Interrupting Collagen Growth (actually a hormone)


  • PGE 2 (Prostaglandin E2) is linked to the onset of and promotion of skin cancer, poor wound healing, and scarring.



One of the prostaglandins, a group of hormone-like substances that participate in a wide range of body functions such as the contraction and relaxation of smooth muscle, the dilation and constriction of blood vessels, the control of blood pressure, and the modulation of inflammation. Prostaglandin E2 (PGE-2) is released by blood vessel walls in response to infection or inflammation and acts on the brain to induce fever. The enzyme PGES-1 is involved in the production of PGE-2 and is an important "switch" for activating the fever response.
